    Setting up and modelling of overflowing fed-batch cultures of Bacillus subtilis for the production and continuous removal of lipopeptides

    This work is related to the setup of overflowing exponential fed-batch cultures (O-EFBC) derived from carbon limited EFBC dedicated to the production of mycosubtilin, an antifungal lipopeptide belonging to the iturin family. O-EFBC permits the continuous removal of the product from the bioreactor achieving a complete extraction of mycosubtilin. This paper also provides a dynamical Monod-based growth model of this process that is accurate enough to simulate the evolution of the specific growth rate and to correlate it to the mycosubtilin specific productivity. Two particular and dependant phenomena related to the foam overflow are taken into account by the model: the outgoing flow rate of a broth volume and the loss of biomass. Interestingly, the biomass concentration in the foam was found to be lower than the biomass concentration in the bioreactor relating this process to a recycling one. Parameters of this model are the growth yield on substrate and the maximal specific growth rate estimated from experiments led at feed rates of 0.062, 0.071 and 0.086 h --1. The model was extrapolated to five additional experiments carried out at feed rates of 0.008, 0.022, 0.040, 0.042 and 0.062 h --1 enabling the correlation of the mean specific growth rates with productivity results. Finally, a feed rate of 0.086 h --1 corresponding to a mean specific growth rate of 0.070 h --1 allowed a specific productivity of 1.27 mg of mycosubtilin g --1 of dried biomass h --1

    Identification and thermochemical analysis of high-lignin feedstocks for biofuel and biochemical production

    Background - Lignin is a highly abundant biopolymer synthesized by plants as a complex component of plant secondary cell walls. Efforts to utilize lignin-based bioproducts are needed. Results - Herein we identify and characterize the composition and pyrolytic deconstruction characteristics of high-lignin feedstocks. Feedstocks displaying the highest levels of lignin were identified as drupe endocarp biomass arising as agricultural waste from horticultural crops. By performing pyrolysis coupled to gas chromatography-mass spectrometry, we characterized lignin-derived deconstruction products from endocarp biomass and compared these with switchgrass. By comparing individual pyrolytic products, we document higher amounts of acetic acid, 1-hydroxy-2-propanone, acetone and furfural in switchgrass compared to endocarp tissue, which is consistent with high holocellulose relative to lignin. By contrast, greater yields of lignin-based pyrolytic products such as phenol, 2-methoxyphenol, 2-methylphenol, 2-methoxy-4-methylphenol and 4-ethyl-2-methoxyphenol arising from drupe endocarp tissue are documented. Conclusions - Differences in product yield, thermal decomposition rates and molecular species distribution among the feedstocks illustrate the potential of high-lignin endocarp feedstocks to generate valuable chemicals by thermochemical deconstruction

    Dose finding and O6-alkylguanine-DNA alkyltransferase study of cisplatin combined with temozolomide in paediatric solid malignancies

    Cisplatin may have additive activity with temozolomide due to ablation of the DNA repair protein O6-alkylguanine-DNA alkyltransferase (MGMT). This phase I/II study determined recommended combination doses using the Continual Reassessment Method, toxicities and antitumour activity in paediatric patients, and evaluated MGMT in peripheral blood mononuclear cells (PBMCs) in order to correlate with haematological toxicity. In total, 39 patients with refractory or recurrent solid tumours (median age ∌13 years; 14 pretreated with high-dose chemotherapy, craniospinal irradiation, or having bone marrow involvement) were treated with cisplatin, followed the next day by oral temozolomide for 5 days every 4 weeks at dose levels 80 mg m−2/150 mg m−2 day−1, 80/200, and 100/200, respectively. A total of 38 patients receiving 113 cycles (median 2, range 1–7) were evaluable for toxicity. Dose-limiting toxicity was haematological in all but one case. Treatment-related toxicities were thrombocytopenia, neutropenia, nausea-vomiting, asthenia. Hearing loss was experienced in five patients with prior irradiation to the brain stem or posterior fossa. Partial responses were observed in two malignant glioma, one brain stem glioma, and two neuroblastoma. Median MGMT activity in PBMCs decreased after 5 days of temozolomide treatment: low MGMT activity correlated with increased severity of thrombocytopenia. Cisplatin–temozolomide combinations are well tolerated without additional toxicity to single-agent treatments; the recommended phase II dosage is 80 mg m−2 cisplatin and 150 mg m−2 × 5 temozolomide in heavily treated, and 200 mg m−2 × 5 temozolomide in less-heavily pretreated children

    Real time in situ microscopy for animal cell-concentration monitoring during high density culture in bioreactor

    International audienceAn in situ microscope (ISM) device is utilised in this study to monitor hybridoma cells concentration in a stirred bioreactor. It generates images by using pulsed illumination of the liquid broth synchronised with the camera frame generation to avoid blur from the cell's motion. An appropriate image processing isolates the sharp objects from the blurred ones that are far from the focal plane. As image processing involves several parameters, this paper focuses on the robustness of the results of the cells counting. This stage determines the applicability of the measuring device and has seldom been tackled in the presentations of ISM devices. Calibration is secondly performed for assessing the cell-concentration from the cell automated numeration provided by the ISM. Flow cytometry and hemacytometer chamber were used as reference analytical methods. These measures and the output of the image processing allow estimating a single calibration parameter: the reference volume per image equal to 1.08 × 10 −6 mL. In these conditions, the correlation coefficient between both reference and ISM data sets becomes equal to 0.99. A saturation of this system during an ultrasonic wave perfusion phase that deeply changes the culture conditions is observed and discussed.     SCA0PEST, a pesticide-free agroforestry cropping system: ex-ante performance evaluation

    From mid 2000, in order to address global challenges, European member states’ strategies for agriculture have progressively identified and given priority to the set up of a multi-functional farm model. In France, first reflection officially started in 2007 through the Grenelle de l'Environnement initiative from which several priority themes were identified, and dedicated measures approved. Concerning agriculture, four different objectives were selected: (a) the deployment of organic farming up to 20 % of the utilized agricultural area by 2020, (b) the Ecophyto plan for a 50 % reduction of the pesticides use, (c) the enforcement of the High Nature Value (HNV) certification scheme and the certification of at least 50 % of the French farms by 2012, and (d) the energetic diagnosis of at least 100.000 farms every 5 years within the farms' energetic performances (PPE) plan. If most of the quantified objectives have not been reached by 2014, coming into force of these measures actually goes on and represents a real opportunity for research organisms to participate to both food security and socio-environmental sustainability by the design, test and deployment of new Productive and Efficient Cropping Systems (PECS). Conversely to the majority of the PECS tested within the EXPE Dephy Ecophyto program, some have been imagined to go further the sole re-conquest of water quality by reducing drastically, or even renouncing chemical pesticides. They actually target (i) the reduction of the farm carbon footprint by reducing and/or mitigating GHG emission, (ii) the improvement of local ecological connectivity by contributing to the establishment emission, (ii) the improvement of local ecological connectivity by contributing to the establishment of new biocorridors and (iii) the financial sustainability of the experimental farms. Among the forty-one EXPE projects funded in 2012 and 2013, only one is testing and monitoring a pesticide-free agroforestry cropping system: SCA0PEST. It aims to: run an experts' panel conceptualization of the SCA0PEST cropping system by considering locally the actual and future market outlets, the hosting parcel and farmhouse agrotechnical potential, and local epidemiologic pressures; to deploy and conduce in farm conditions the cropping system; assess global and thematic performances of it; and favour the disclosure of the obtained knowledge and references towards farmers, advisers and agricultural student
